Genetic disruption of Tall (Scl) and Lmo2 individually results in defective yolk sac erythropoiesis and definitive hematopoiesis. These transcription factors physically interact with each other to form an Lmo2/Tall/Gata1/2/3 transcription factor complex. In order to know whether these transcription factors form a complex to play critical roles in hematopoiesis, mutant Lmo2s were made to specifically disrupt the interaction between Lmo2 and Gata1/2/3 by introducing a mutation in the LIM 2 domain of Lmo2. Overexpression of these mutant Lmo2s in mice severely impaired fetal liver hematopoiesis and thymus development, indicating its role as a master complex for the development of the entire hematopoietic system.
